When I look at contest exchanges, it says serial number.
Does that mean I tell them the number of contact they are?
So I would say to my first contact "Your 01"
Am I correct?

You got it! For those contests requiring a serial number, the exchange for the 1st QSO in the contest would be '1' plus whatever other info is required per the various rules.
(Well, technically, it would be "You're 01". But I digress :P)
