When you start whith a contest you will start with the 001 number and then 002 and so on.
By example
cq contest sv5frv cq contest
and the you take the microfone
sv5frv wc2hh you signal report is 59 and your serial number is 001
and after i will tell you wc2hh sv5frv thanks for 001 your number is by example 152

The numbers mean the number of the qso's that you have done

If i am mistaken i pleaze all the other radio amateurs to correct me
Hope i have helped you

You may have been on the air during the wpx ssb contest last month. In that contest serial numbers are used.
Not all contests use serial numbers.
Just to list a few that do not:
North America QSO Party
IARU HF World Championship
And a few that do:
North America Sprint (qso #, name, state)
Check out the rules of any contest you may want to play in so that you will know what the exchange is supposed to be and it will give you a better understanding of the concept of a given contest.
